چکیده

Specific objectives of this research include the following: 1) Measurements of inherent and apparent optical properties will be conducted in areas frequented by Karenia brevis bloom events off the west Florida coast, and attempt to identify key optical indices for discriminating blooms of K. brevis from other phytoplankton and other sources of optical variability. 2) We will examine the utility of inversion methods to retrieve phytoplankton absorption from reflectance, and subsequently evaluate this with regard to optical criteria for detection of K. brevis and other phytoplankton taxa.

Use of hyperspectral remote sensing reflectance for detection and assessment of the harmful alga, Karenia brevis.

We applied two numerical methods to in situ hyperspectral measurements of remote sensing reflectance Rrs to assess the feasibility of remote detection and monitoring of the toxic dinoflagellate, Karenia brevis, which has been shown to exhibit unique absorption properties. First, an existing quasi-analytical algorithm was used to invert remote sensing reflectance spectra, Rrs(lambda), to derive ...

Progress in developing a new detection method for the harmful algal bloom species, Karenia brevis, through multiwavelength spectroscopy

Multiwavelength spectroscopy is a rapid analytical technique that can be applied to detect, identify, and quantify microorganisms such as Karenia brevis, the species known for frequent red-tide blooms in Florida’s coastal waters. This research will report on a model-based interpretation of UV–vis spectra of K. brevis. The spectroscopy models are based on light scattering and absorption theories...

Temporal variation of caspase 3-like protein activity in cultures of the harmful dinoflagellates Karenia brevis and Karenia mikimotoi

To better understand the role of caspase-like proteins in the cellular activities of harmful dinoflagellates, cultures of Karenia brevis and Karenia mikimotoi were monitored over a 56-day period. The cellular abundance, maximum photochemical efficiency of photosystem II (Fv/Fm), percentage of dead cells (revealed by SYTOX-labelling of nuclei of cells with a compromised membrane) and the variati...

Vertical Migration of the Toxic Dinoflagellate Karenia Brevis and the Impact on Ocean Optical Properties

Vertical migration behavior is found in many harmful algal blooms; however, the corresponding impact on ocean optical properties has not been quantified. A near-monospecific population of the dinoflagellate Karenia brevis was encountered off the west coast of Florida. The community was tracked for 24 hours by following a Lagrangian drifter deployed at the beginning of the experiment. A suite of...

Extraction and analysis of lipophilic brevetoxins from the red tide dinoflagellate Karenia brevis.

Efficient extraction and accurate analysis of lipophilic brevetoxins (PbTxs), produced by the harmful algal bloom (HAB) species Karenia brevis, are essential when assessing the toxicological potential of this dinoflagellate. One of the most commonly used brevetoxin extraction methodologies employs C18 solid-phase extraction (SPE). In this study, C18 SPEC discs were tested for extraction of spik...
